Automated Coupled Ion ExclusionIon Chromatography for
the Determination of Trace Anions in Fermentation Broth

Although conventional ion chromatography is well-suited for the determination of trace inorganic anions, it suffers from a matrix interference if the aqueous sample contains a relatively high concentration of weak carboxylic acids (organic acids). Attempts to measure chloride, nitrate, and sulfate ions at parts-per-million concentration levels in sample matrices that have a 10% or higher organic acid content are impossible. Combining ion-exclusion chromatography with low capacity anion exchange (IEIC) solves the organic acid matrix problem and has wide utility to problems encountered in applying ion chromatography to biotechnology. A Dionex BioLC ion chromatograph is modified so as to accomplish IEIC. The modified instrument is connected to a dual channel interface that affords simultaneous dual ion chromatograms. To achieve acceptable precision and accuracy, an internal standard mode of calibration had to be developed. This paper discusses our experience with IEIC and shows our success on actual pilot plant samples from a proprietary biotechnology process.
